产品背景介绍：
G 蛋白偶联受体激酶( G protein-coupled receptor kinases , GRKs) 是一簇与G蛋白偶联受体( GPCRs) 快速失敏( desensitization) 相关的激酶。许多GPCRs 如阿-片受体、血栓素受体、52羟色胺受体、肾上腺素能受体等在激动剂持续刺激时易发生转导信号的快速衰减,这种调节机制主要与GRKs有关。GRKs 家族由7 个结构上有同源序列的家族成员组成。每种GRKs 都含有共同的功能结构,包括1个中心催化区、1 个底物识别和含有G蛋白信号调节蛋白( regulators of Gprotein signaling ,RGS) 样结构的氨基末端区,以及1 个作用于胞膜的羧基末端区。
根据序列和功能的相似性可分为3 个亚家族：
第一个亚家族包括GRK1 和GRK7 。GRK1 是视紫质( rhodopsin) 激酶,仅在视网膜光受体细胞表达,作用底物是视网膜的视蛋白。
第二个亚家族的GRK2 和GRK3 又分别称为β2肾上腺素能受体激酶1 (β2ARK1) 和β2肾上腺素能受体激酶2 (β2ARK2)。
第三个亚家族包括GRK4 , GRK5 和GRK6 。
GRK4 仅在睾丸高水平表达,提示该酶具有底物的特异性。
G protein-coupled receptor kinases (GRKs, GPCRKs) are a family of protein kinases that regulate the activity of G protein- coupled receptors (GPCRs) by phosphorylating their intracellular domains after their associated G proteins have been released and activated. The phosphorylated serine and threonine residues act as binding sites for arrestin proteins that prevent the reassociation of the G proteins with their receptors, thereby preventing reactivation of the signaling pathway. GRKs regulate also cellular responses independent of their kinase activity. In particular, G protein-coupled receptor kinase 2 interacts with a diverse repertoire of non-GPCR substrates.
